Virtual EventTitle: High-throughput Studies of Macromolecular Complexes using Flow Magnetic Tweezers
Abstract: The application of forces and torques on a single-molecule level has transformed our understanding of the dynamic properties of biomolecules, but limited throughput of these techniques has hindered the characterisation of rare events and intrinsic heterogeneity in a statistically robust manner. Here, we introduce a multiplexed single-molecule technique called flow magnetic tweezers (FMT) that provides a 100-fold improvement in throughput of force spectroscopy measurements with topological control. Using FMT, we characterized the supercoiling dynamics and drug-induced DNA break intermediates of E. coli DNA gyrase as well as screen diverse topoisomerases. The method and the accompanying software with an automated feature classification pipeline are used to study a broad range of enzymes and macromolecular complexes (like replisomes) involved in nucleic acid transactions. FMT also shows promise as an antibiotic drug-screening platform.
