DESCRIPTION:Proteins carry out the majority of cellular functions. While si
 ngle-cell transcriptomics provides rich information on the expression level
  of proteins\, it does not reveal the entire cellular proteome due to trans
 criptional regulation and post-translational modifications. As a result\, t
 he consequences of changes in expression can only be fully understood by as
 sessing the protein content of a cell. This has spurred considerable intere
 st in the development of technologies that enable single-molecule analysis 
 and quantitation of proteins. Although important headway is being made rega
 rding single-molecule protein profiling and quantitation\, there is still a
 n unmet need for methods that can accurately identify a wide range of prote
 ins—including PTM variants—at a meaningful throughput. In this seminar\, I 
 will present my research on the development of peptide sequencing via chemo
 enzymatic “reverse translation” of peptide sequences into barcoded DNA libr
 aries that encode the sequence of peptides. This approach\, for the first t
 ime\, enables peptide sequence information to be amplified by PCR and analy
 zed via high-throughput DNA sequencing. With further refinement and develop
 ment\, this approach will be amenable to the massively parallel profiling o
 f both unmodified and PTM-bearing proteins\, opening up exciting new opport
 unities for single-molecule proteomics.
