Single Molecule Visualization of Rad54 and Rdh54 Organization During Homologous Recombination

DNA motor proteins control all aspects of nucleic acid metabolism. Including DNA replication, recombination, and repair.  Involvement of DNA motor proteins in these diverse, yet related, pathways provides an opportunity to discover key aspects of basic DNA biology. In my research group we use single molecule optical microscopy to directly visualize the organization and dynamics of DNA motor proteins in real-time. Our focus is to understand how these proteins function to prevent the development of human cancers. Today, I will discuss some of my recent work on the structural and functional organization of two highly related Snf2 DNA motor proteins, Rad54 and Rdh54, within key homologous recombination intermediates.  Findings from these studies shed new light on how these two motor proteins function during DNA repair.
