Mechanisms of Aggregation of the Mammalian Prion Protein
The misfolding and aggregation of the prion protein is associated with several neurodegenerative diseases. To understand how these processes begin, has been a long-standing challenge. Using the tools of molecular biology, physical biochemistry and spectroscopy, the structural perturbations that occur in the monomeric protein prior to oligomerization have been identified, and insight has been obtained into the sequence of the major conformational conversion steps that occur in the oligomeric state.
